Protein quantification
We observed that the four extraction buffers rendered significantly
different protein concentrations for peanuts (Figure 1). The highest CPE
yield was obtained with BB2ßME (22,82 ± 1,19 mg/mL) followed by SB
(20,94 ± 2,33 mg/mL), BB (18,72 ± 2,92 mg/mL) and TB/HCL (12,99 ± 2,32
mg/mL). These results show the different extraction potential of each
buffer. Considering the use of 10 grams of the seed and 100 mL of each
extraction buffer the final CPE yield was as follows: BB2ßME (62.9%);
SB (59.3%), BB (51,6%) and TB/HCL (38.2%). The final volume obtained
of each extract did not significantly influence the total protein
extraction BB2ßME (75mL); SB (77mL), BB (75mL) and TB/HCL (80mL).
